“Seedling is coming”: highlighting novel players in the seed pre-germinative metabolism

The enhancement of seed quality is crucial for the majority of farming systems. Seed priming comprises a range of pre-sowing treatments based on an incomplete imbibition that activates pre-germinative metabolism, thus preparing the seed for a faster germination, a more uniform seedling establishment and an improved stress tolerance. Primed seeds are subsequently dehydrated and stored before sowing, but prolonged priming impairs desiccation tolerance and seed viability, a condition known as “overpriming”. Since rehydration and dehydration trigger ROS production and genotoxic stress, the maintenance of genome integrity plays a major role in desiccation tolerance, although the interaction between DNA Damage Response, antioxidant response and desiccation tolerance remains unclear in the context of seed priming protocols. The research exposed in this work aims to address this gap of knowledge using primed versus overprimed seeds of the model legume Medicago truncatula.
Biometric analyses of germination rates and seedling development allowed the determination of priming and overpriming conditions, whereas a boost in ROS production, accumulation of DNA lesions, alterations in nucleolar morphology and the differential regulation of genes involved into DNA repair and antioxidant response were detected in overprimed seeds at different stages of post-priming dehydration. This integrative approach aims to deliver molecular hallmarks of desiccation tolerance and priming efficacy, providing novel insights for the optimization of seed priming protocols.
